The key (proteins) to self-renewing skin

ScienceDaily (July 5, 2012) ? In the July 6 issue of Cell Stem Cell, researchers at the University of California, San Diego School of Medicine describe how human epidermal progenitor cells and stem cells control transcription factors to avoid premature differentiation, preserving their ability to produce new skin cells throughout life.

The findings provide new insights into the role and importance of exosomes and their targeted gene transcripts, and may help point the way to new drugs or therapies for not just skin diseases, but other disorders in which stem and progenitor cell populations are affected.

Stem cells, of course, are specialized cells capable of endlessly replicating to become any type of cell needed, a process known as differentiation. Progenitor cells are more limited, typically differentiating into a specific type of cell and able to divide only a fixed number of times.

Throughout life, human skin self-renews. Progenitor and stem cells deep in the epidermis constantly produce new skin cells called keratinocytes that gradually rise to the surface where they will be sloughed off. One of the ways that stem and progenitor cells maintain internal health during their lives is through the exosome -- a collection of approximately 11 proteins responsible for degrading and recycling different RNA elements, such as messenger RNA that wear out or that contain errors resulting in the translation of dysfunctional proteins which could potentially be deleterious to the cell.

"In short," said George L. Sen, PhD, assistant professor of medicine and cellular and molecular medicine, "the exosome functions as a surveillance system in cells to regulate the normal turnover of RNAs as well as to destroy RNAs with errors in them."

Sen and colleagues Devendra S. Mistry, PhD, a postdoctoral research fellow, and staff scientist Yifang Chen, MD, PhD, discovered that in the epidermis the exosome functions to target and destroy mRNAs that encode for transcription factors that induce differentiation. Specifically, they found that the exosome degrades a transcription factor called GRHL3 in epidermal progenitor cells, keeping the latter undifferentiated. Upon receiving differentiation inducing signals, the progenitor cells lose expression of certain subunits of the exosome which leads to higher levels of GRHL3 protein. This increase in GRHL3 levels promotes the differentiation of the progenitor cells.

"Without a functioning exosome in progenitor cells," said Sen, "the progenitor cells prematurely differentiate due to increased levels of GRHL3 resulting in loss of epidermal tissue over time."

Sen said the findings could have particular relevance if future research determines that mutations in exosome genes are linked to skin disorders or other diseases. "Recently there was a study showing that recessive mutations in a subunit of the exosome complex can lead to pontocerebellar hypoplasia, a rare neurological disorder characterized by impaired development or atrophy of parts of the brain," said Sen. "This may potentially be due to loss of progenitor cells. Once mutations in exosome complex genes are identified in either skin diseases or other diseases like pontocerebellar hypoplasia, it may be possible to design drugs targeting these defects."

Centro Fund Services, a joint venture between IDS Group and Point Nine, announced the launch of its independent middle office services to the South African investment manager market with the opening of an office in Cape Town.

In a statement, Centro, which means middle in Italian, said that its independent middle office services provide day to day portfolio administration covering trade capture, counterparty trade reconciliations, settlements, reconciliations with other third parties such as fund administrators, prime brokers and custodians, administering corporate actions and delivering related reports. The new service will use Point Nines proprietary web-based platform, Circle. Traditionally South African investment managers use in-house teams to carry out their middle office services. But this practice has become too costly and complex to manage notwithstanding the ongoing international trend to outsource such services to a reputable service provider who is able to offer efficient and cost saving services based upon economies of scale and excellent software.

While Google may have grabbed headlines for its recent wearable tech, Olympus is doggedly forging ahead with its own prototypes, seven years on. Unlike Project Glass, the MEG4.0 isn't a standalone structure and needs a glasses frame to hang on, although the sub-30g unit shouldn't tax it too much. The QVGA (320 x 240) display can connect to devices through Bluetooth 2.1, with Olympus pointing to a smartphone hook-up to provide both the processing power and internet connectivity -- which sounds different to what we're expecting from Google's effort. The current prototype can squeeze out eight hours of intermittent use, or two hours of non-stop projection. While the device is being pitched at everyday users, Olympus aren't offering any suggestion at launch dates or pricing, but you can check on what the company is willing to share in the (Google-translated) press release below.

AIB will not write down any mortgage debt as part of its plans to tackle arrears, an internal memo has revealed.

The bank outlined four specific schemes which it is to roll out for existing primary residential mortgage customers, according to head of mortgages Jim O?Keeffe. These include:

* Standard/short-term forbearance: Where the difficulty is deemed to be short-term, the solution could be interest only, fixed repayment, repayment break, extension of the loan term, capitalisation of arrears, or deferred interest scheme;

* Split mortgage: This will be considered where a customer can afford a mortgage but their income is not sufficient to fully support it. The mortgage is split into two parts. Loan A, repaid on the basis of capital and interest, and Loan B, which is deferred and becomes repayable at a later date;

* A negative equity trade-down: A customer will reduce monthly loan repayments and overall indebtedness by trading down to a property more appropriate to individual circumstances. Following the trade-down, the maximum loan to value must not exceed 175%, inclusive of the negative equity amount being carried forward from the previous property;

* Voluntary sale for loss: This will be considered where the loan is deemed to be unsustainable and the customer is agreeable to sell the property and put an appropriate agreement in place to repay residual debt.

The memo said further details on the buy-to-let strategy would be communicated separately.

The number of private residential mortgages in arrears of 90 days or more at AIB have risen from 6.9% at the end of Q1 2011 to 7.7% at the end of Q1 2012.

Following publication of the Personal Insolvency Bill, there had been hope that mortgage holders in negative equity would be able to write off some of their debts by cutting deals with banks by threatening to file for bankruptcy. However, director of the Free Legal Advice Centre Noeline Blackwell said she was not surprised AIB was not offering a writedown on mortgage debt.

The concern for Flac is, if a bank owns 65% of a person?s debt, they can block them from pursuing personal insolvency arrangements.
